Nuclear Shaping in the Absence of Microtubules in Scorpion Spermatids
نویسنده
چکیده
During spermiogenesis in most animal species, sperm nuclei undergo morphogenesis which results in the transformation of relatively large spherical nuclei with diffuse chromatin into small elongate or flattened nuclei with highly condensed chromatin. The shapes of nuclei of mature spermatozoa are beutifully precise and species-specific. There is no general agreement among cell biologists as to the forces which effect nuclear shaping. Some investigators have suggested that microtubules which surround spermatid nuclei during shaping play an active role in this process (Mclntosh and Porter, 1967; Kessel, 1966, 1970), whereas others have suggested that nuclear condensation is effected by internal condensation of chromatin (Fawcett et al., 1971). It has also been suggested that nuclear condensation and shaping could result from the interplay of nucleus, cytoplasmic organelles, and supportive cells (Phillips, 1970; Rattner, 1972). In this report, I shall describe nuclear shaping during spermiogenesis in two species of scorpion where the formation of a highly compacted sperm nucleus from a large spherical nucleus appears to be effected without interaction with microtubules.

Actin related protein of vertebrate, Arp1, is a major component of the dynactin complex. To characterise and localise Arp1 during mammalian spermatogenesis, polyclonal antibodies were raised against a human recombinant Arp1. Anti-Arp1 antibodies were used for western-immunoblotting, indirect immunofluorescence and immunoelectron microscopy. In round spermatids, Arp1 was detected at the centroso...
متن کاملKIFC1 is essential for acrosome formation and nuclear shaping during spermiogenesis in the lobster Procambarus clarkii
In order to study the function of kinesin-14 motor protein KIFC1 during spermatogenesis of Procambarus clarkii, the full length of kifc1 was cloned from testes cDNA using Rapid-Amplification of cDNA Ends (RACE). The deduced KIFC1 protein sequence showed the highest similarity between Procambarus clarkii and Eriocheir senensis (similarity rate as 64%). According to the results of in situ hybridi...
متن کاملKIFC1-Like Motor Protein Associates with the Cephalopod Manchette and Participates in Sperm Nuclear Morphogenesis in Octopus tankahkeei
BACKGROUND Nuclear morphogenesis is one of the most fundamental cellular transformations taking place during spermatogenesis. In rodents, a microtubule-based perinuclear structure, the manchette, and a C-terminal kinesin motor KIFC1 are believed to play crucial roles in this process. Spermatogenesis in Octopus tankahkeei is a good model system to explore whether evolution has created a cephalop...
متن کاملPreparation and biodistribution study of a 99mTc-labeled toxic fraction of Iranian mesobuthus eupeus scorpion venom
Introduction: Iranian scorpion species are classified in Buthidae and Scorpionidae with 16 genera and 25 species. In Iran, similar to other parts of the world, there are a few known species of scorpions responsible for severe envenoming. Mesobuthus eupeus is the most common species in Iran. Its venom contains several toxin fractions which can affect the ion...
متن کاملP-121: Assessment of Microtubule and Nuclear Status at Different Intervals of Bovine In Vitro Oocyte Maturation
Background: Mammalian oocyte undergoes a series of structural nuclear modulations during maturation in order to obtain full competence to support fertilization and early embryonic development. Microtubules are major cytoskeleton components and have pivotal modulators for chromosomal movement, and it has been shown that partial or immature spindle organization may compromise correct ploidy of th...
